SUMMARY:סמינר אורח: ד"ר יעל דוד
DESCRIPTION:Metabolism Meets Epigenetics: Chemical Biology in Translational
Discovery\n ** Lecture will be given in English**\nAbstract:\nThe epigen
etic regulation of DNA-templated processes plays a fundamental role in sha
ping cell identity and fate. Disruption of these regulatory networks contr
ibutes to numerous diseases\, including cancer\, viral infection\, neurona
l function and immune dysfunction. Many of these pathologies are accompani
ed\nby profound alterations in genome organization\, including the formati
on of non-canonical chromatin structures. Our work has uncovered the epige
netic regulation of several such structures\, including micronuclei (Natur
 e\, 2023)\, extracellular vesicle-associated chromatin (Nature Cancer\, 20
 24)\, and the\nchromatinized genome of Hepatitis B virus (HBV) (Cell\, 202
5). By investigating the molecular mechanisms underlying these processes\,
we identified cellular metabolism as a major regulator of chromatin funct
ion. We found that intrinsic metabolic programs\, as well as environmental
influences such\nas nutrition and diabetes\, directly reshape the chromat
in landscape through chemical modifications \, thereby altering gene expre
ssion and cell fate (Nature Communications\, 2019\; Nature Communications\
, 2020 .)In this talk\, I will present our latest discoveries defining thi
s metabolism-epigenetics axis and\ndiscuss how metabolic stress reprograms
chromatin to regulate T cell function. I will also highlight how these me
chanistic insights are being translated into the development of a new gene
ration of CAR T cells with enhanced therapeutic potential. Together\, thes
e studies illustrate how fundamental discoveries in\nchemical biology and
epigenetics can reveal unexpected mechanisms of disease and inspire innova
 tive therapeutic strategies.
