SUMMARY:Biomolecular Recognition &amp\; Protein Evolution
DESCRIPTION:Biomolecular recognition is fundamental to the cellular functio
 ns via the formation of binding complexes. Functional complex of biomolecu
 lar recognition normally has high affinity and specificity which are shape
 d by the evolution. The affinity determines the stability of the complex w
 hile the specificity determines the discrimination of native complex again
 st competitive ones. The affinity can be quantified computationally and ex
 perimentally. However\, the specificity is challenging to be quantified si
 nce the comparison of all possible binding partners for the target biomole
 cule is a daunting task.\n\nBiological activities are mostly performed by 
 the proteins at the molecular scale. Naturally occurring proteins have a h
 igh degree of features different from random mixer of amino acids. They ha
 ve evolved to be able to spontaneously folding into native structure and s
 pecifically bind with their partners for functions. Darwin’s Theory of E
 volution suggests that natural selection imposes on the individuals and th
 e fittest survives. A fundamental question is how nature shapes the protei
 n interaction patterns for stable structure and specific functional bindin
 g at the molecular scale.\n\nIn this lecture\, I will introduce these two 
 topics (biomolecular recognition &amp\; protein evolution) and the attempt
  to address the raised challenge and question with physical concept\, math
 ematical statistics and computational approaches. I will focus on the deve
 lopment of the scoring function based on both the affinity and specificity
  optimizations\, and the unification of physical principles for protein fo
 lding\, binding and evolution.

DESCRIPTION:Molecular diagnostics have the potential to revolutionize our a
 bility to diagnose\, monitor and treat a variety of diseases. However\, de
 tecting circulating biomarkers has proven to be challenging due to the low
  concentrations of many biomarkers. Furthermore\, other factors including 
 sample heterogeneity\, low sample volume\, and cross-reactivity between di
 fferent markers complicate biomarkers detection. Consequentially\, there i
 s a need for improved biomarker detection methods.  The emergence of sing
 le-molecule techniques opens new opportunities to overcome these challenge
 s. Counting individual molecules enables the detection of biomarkers at lo
 w quantities. Additionally\, single-molecule technologies are particularly
  relevant to the analysis of heterogeneous samples\, and can allow for bio
 markers detection with a wide dynamic range. The Walt lab previously devel
 oped an ultra-sensitive assay known as Single Molecule Arrays (Simoa) by i
 solating single molecules into femtoliter-sized compartments using microfl
 uidics\, and detecting them optically. The Simoa technology enables biomar
 kers to be measured at concentrations a thousand times lower than current 
 molecular diagnostic methods\, providing a more accurate approach for diag
 nosing certain forms of cancer\, infectious diseases and neurodegenerative
  disorders.  Recently\, the COVID-19 pandemic has led to an urgent need f
 or robust diagnostic assays. To address this need\, we developed ultra-sen
 sitive Simoa based methods for the detection of anti-SARS-CoV-2 antibodies
  and viral antigens\, and an assay for the quantification of antibody neut
 ralization capacity. We used these new assays to study important clinical 
 problems in collaboration with clinicians in Brigham and Women's Hospital 
 and Massachusetts General Hospital. For example\, we studied the mechanism
  of Multisystem Inflammatory Syndrome in Children (MIS-C). Our new finding
 s on MIS-C pathogenesis provide novel targets for treating and preventing 
 MIS-C\, which are urgently needed for this increasingly common\, severe CO
 VID-19-related disease in children. The past year has expedited the use of
  Simoa in the clinic\, and we are now excited about further developing and
  applying single molecule detection methods for numerous clinical applicat
 ions.\n\n&nbsp\;\n\nAbstract
