Research Topic: gainsta Derived Peptide-Antimicrobial Activity of URS Klebsiella pneumoniae

The increasing prevalence of antibiotic-resistant Klebsiella pneumoniae highlights the need for novel antimicrobial strategies. This study investigated the antibacterial potential of peptides derived from underrepresented sequences (URSs), examining both extracellular application and intracellular expression. For this purpose, a stable inducible expression system was first developed in K. pneumoniae to enable controlled intracellular peptide expression.
The antimicrobial activity of the URS-derived peptide RLLRRCMYW was evaluated against capsulated and non-capsulated K. pneumoniae, both alone and in combination with conventional antibiotics. In parallel, an inducible expression system was developed to investigate the effects of intracellular expression of CMYW-based peptides. Extracellular application of the peptide inhibited bacterial growth, while combination treatments produced greater inhibition than either treatment alone. Intracellular expression of CMYW-based peptides resulted in a substantial reduction in bacterial viability .
These findings demonstrate the antimicrobial potential of URS-derived peptides and support further investigation of their mechanisms of action and development as a novel class of antimicrobial agents.
